Abstract

Objective: To observe the effects of Shen Tong Zhu Yu decoction on the proliferation of osteoarthritis (OA) synovial fibroblasts and expression of cyclooxygenase-2 (COX-2). Methods: Synovial fibroblasts were separated and identified by flow cytometry. Synovial fibroblasts were cultured in saline (negative control), etoricoxib containing serum (positive control), and Shen Tong Zhu Yu decoction contained serum. The proliferation ability of synovial fibroblasts was detected by MTT. The expression of COX-2 was detected by RT-PCR and western blot. Results: Compared with a negative control, etoricoxib containing serum and Shen Tong Zhu Yu decoction containing serum both can inhibit the proliferation ability of synovial fibroblasts (P<0.05), and there was no significant difference between etoricoxib containing serum and Shen Tong Zhu Yu decoction contained serum (P>0.05). RT-PCR and western blot shows that etoricoxib containing serum and Shen Tong Zhu Yu decoction containing serum both can inhibit the expression of COX-2, and there was no significant difference between etoricoxib containing serum and Shen Tong Zhu Yu decoction containing serum (P>0.05). Conclusion: Shen Tong Zhu Yu decoction containing serum can inhibit the proliferation ability of synovial fibroblasts and the expression of COX-2, which maybe one of the potential mechanisms for the treatment of OA.
